The degree of protein enrichment of sweet potato residue by different amylolytic moulds in solid-state cultivation was much greater than that obtained using amylolytic yeasts. The optimum initial moisture content for protein enrichment was about 65% (w/w). Adding nitrogen sources to the culture twice (at the start of the incubation and after 24 h) considerably improved the final protein content. A co-culture of amylolytic mycelial fungi yielded a product with 32% (w/w) crude protein after 4 days' incubation at 30°C. In a column reactor, the highest temperatures reached were 42°C and 40°C and the minimum O2 concentrations were 1.5% and 2.5% of full saturation in the central and bottom layers, respectively. 相似文献
Arbuscular mycorrhizal (AM) fungi can form symbiosis with 90% of the vascular plants and play important roles in ecosystem. To realize the AM fungal colonization at different succession stages in saline-alkali land and screen AM fungi species with great functions, roots and soil samples were collected from the three succession stages of Songnen saline-alkali grassland. The soil properties and AM fungal colonization were measured, and the fungus distributed extensively in three stages was annotated by sequencing for AML1/AML2 target, subsequently, maize was selected as the host to verify its colonization. The results showed that the soil properties improved with the succession of saline-alkali grassland. The plants’ communities of the three stages could be colonized by AM fungi, and the colonization rate of Leymus chinensis (the third stage) ranged from 66.67% to 100%, Puccinellia tenuiflora (the second stage) ranged from 50% to 80%, while the Suaeda glauca (the first stage) was only 35%–60%. Glomeraceae sp1 was identified as the dominant AM fungi species which occurred frequently in the succession of saline-alkali land with the isolation frequency, relative abundance, and importance value of 100%, 18.1%, and 59.1%, respectively. The colonization rate of Glomeraceae sp1 in maize ranged from 80% to 87% and similar mycorrhizal characteristics were detected in the roots of P. tenuiflora, S. glauca, and L. chinensis, indicating that Glomeraceae sp1 colonized the samples in the field. The correlation matrix indicated that colonization rate, colonization intensity, and vesicle abundance were closely related to soil conditions most, and they were related significantly to all the soil properties except cellulase activity. Besides, redundancy analysis (RDA) showed that soil properties drove the changes of AM fungal colonization and sporulation. These results will provide theoretical support for realizing the relationship between AM fungal colonization and soil conditions, and also for the exploration of AM fungi species with great functions.
【背景】海洋沉积物真菌富含生物活性天然产物,但珊瑚礁泥砂真菌及其天然产物的研究较少。【目的】分离珊瑚礁泥砂真菌及其天然产物,探究珊瑚礁泥砂来源真菌多样性,为海洋真菌天然产物开发奠定基础。【方法】采用稀释涂布平板法分离马来西亚热浪岛珊瑚礁泥砂真菌并基于ITSrDNA序列分析鉴定真菌;综合运用硅胶柱、反相柱和制备HPLC色谱技术分离枝孢属真菌(Cladosporium sp.) GXIMD02067的天然产物,通过核磁共振波谱技术和文献数据比对鉴定化合物结构。【结果】19株真菌被分离,隶属1纲4目4科6属,包括7株曲霉属(Aspergillus)、6株青霉属(Penicillium)、2株枝孢属(Cladosporium)、1株蜡蚧菌属(Lecanicillium)、2株路霉属(Lulworthia)和1株Parengyodontium。GXIMD02065和GXIMD02066 ITS rDNA序列的相似度小于87%,是潜在新菌种。7个化合物从Cladosporium sp. Roots of 28 species of epiphytic vascular plants were collected on tree trunks and branches at six afromontane forest sites between 1700 and 3300 m above sea level in Bale Mountains National Park, Ethiopia. Seven of the 28 epiphyte species were colonized by vesicular-arbuscular mycorrhizal fungi (VAM). Mycorrhizal colonization only occurred at two of the six sites examined, at 2900 m and 3300 m, but more than one type of VAM endophyte was present in each case. Three facultative epiphytic species were all highly colonized by VAM on the forest floor, whereas roots from epiphytic habitats were weakly colonized. No correlations were found between VAM colonization, fine root diameter and root hair length, but VAM colonization and root hair abundance were negatively correlated. The lack of VAM colonization of potential, epiphytic host species at the majority of the sites examined points to the dispersal of VAM propagules as the factor limiting mycorrhizal colonization of epiphytic habitats. It is suggested that root systems of hemiepiphytic tree species serve as corridors between forest floor and tree trunks through which VAM may spread via hyphal growth. 相似文献
The formation of vesicular-arbuscular mycorrhizae (VAM) in intact soil profiles from two sites in southeastern Australia were measured at two depths using a bioassay grown in intact soil cores. Intact soil cores were taken from (1) topsoil (0–15 cm) and (2) subsoil (15–30 cm) four times during 1990. Seeds of Acacialinifolia (Vent.) Willd. (Mimosaceae) were sown into the cores and plants harvested 8 and 12 weeks after sowing. For 1990, at both sites and in all seasons, VAM most readily developed in the roots of seedlings of A. linifolia grown in topsoil. Limited VAM occurred in roots grown in subsoil cores. Most colonisation of roots by VAM occurred from cores collected during spring and summer. Spore numbers were quantified for each site and depth by wet-sieving 100-g samples of air-dried soil and counting turgid spores containing oil droplets. Three types of spores were found in the soils. Few spores were extracted from all soils sampled, and for the most abundant of the spore types at least twice as many spores occurred in the topsoil than in the subsoil for all seasons examined. As most of the propagules that initiate VAM infection were observed in the topsoil, disturbances which involve the removal and storage of the top 15 cm will adversely affected these fungi. 相似文献
